In Reply: Dr Frühbeck and colleagues raise several important points related to leptin physiology as it applies to our study. Our evaluation of weight loss was across the dose range and not just at the higher leptin dose. Also, our study was designed to examine pharmacologic rather than biological effects of leptin. Pharmacokinetic results show that the amount of leptin administered far exceeded endogenous secretion. Additional studies have been conducted, as suggested by Frühbeck et al, evaluating morning vs evening leptin administration and the analysis of these results is ongoing.
